The PF-TEK method is named after a way of cultivating mushrooms, developed by Robert McPherson (also known as Psilocybe Fanaticus) in 1991. It remains a popular and simple technique for growing magic mushrooms such as Psilocybe cubensis. This method uses a substrate consisting of vermiculite, brown rice, and water. The mixture is steamed and kept in sterile jars. Liquid culture is then added with a syringe, and cultivation begins. The technique is loved for its accessibility, affordability, and relatively low chance of contamination. PF-TEK is ideal for beginners and hobbyists in mushroom cultivation looking to go beyond ready-to-grow kits.

How to Cultivate Psilocybe cubensis Mycelium with Agar Agar

  1. Prepare Materials: Ensure you have a sterile workspace, a sterile spore print, a Petri dish, a blade, and distilled water. Work sterilely. Wear gloves and a mask and clean your environment thoroughly.
  2. Prepare Agar Agar Mixture:
    • Determine how much you need. It is better to have slightly more than too little. For a Petri dish, you only need a layer of 1 to 5 millimeters. Make sure the entire bottom is covered. We recommend filling multiple Petri dishes, as even professionals often encounter contamination, thus increasing your chances of success.
    • Mix 1 gram of agar agar powder with 50 ml of distilled water. More can be made at once, but the ratio is 1 gram of powder to 50 ml of water.
    • You can also add half a gram of rye flour to the mixture. These mushrooms particularly like it.
    • Bring the mixture to a boil until the agar is fully dissolved.
  3. Sterilize:
    • Place your Petri dishes or other heat-resistant containers in a pot with a lid, a pressure cooker, or an autoclave and add a small layer of distilled water to the pot.
      1. Regular Pot: Ensure the pot never boils dry. Add enough water. Place the dishes separately on a cloth or rack in the pot. Cooking time: 60 to 90 minutes.
      2. Pressure Cooker and Autoclave: Start the timer once the pressure cooker has reached the desired pressure. After sterilization, allow the pressure to gradually decrease before opening to prevent dishes from cracking due to sudden pressure changes. Cooking time: 20 to 30 minutes at 15 PSI (approximately 1.03 bar).
  4. Pour the Agar:
    • Pour the sterile, warm agar into sterile Petri dishes or culture trays. Ensure the mixture does not cool too quickly. Close the dishes immediately after pouring to prevent contamination. Seal the edges with, for example, parafilm that leaves no adhesive residue.
  5. Inoculate with Spores:
    • Heat a knife tip with a gas stove or a blue-flame lighter. Allow it to cool to room temperature.
    • Only now open the spore print. Do this quickly and purposefully. Scoop a small amount of spores from the spore print and spread them evenly in the Petri dish.

After a few days, white mycelium threads will appear if all steps were performed sterilely and carefully. Regularly check for contamination and remove any affected dishes. Contamination is recognizable by any color other than white. If you have successfully cultivated mycelium, you can transfer this mycelium into a jar with, for example, rye flour, brown rice, or other grains of your choice. Again, work sterilely. Soak the grain in water and sterilize the jars with soaked grain before adding the mycelium. Ensure it can breathe but prevent contamination with, for example, a Teflon sticker over a hole in the lid or microporous tape with holes smaller than 0.3 micrometers.
